When you work with 4 or more columns you have the same decisions as with 2 or 3 columns but you start running into constraints. Monitors might be getting larger, but people are not browsing maximized any longer. So the more columns you have, the more you run the risk that your design won't display correctly or it will force your readers to scroll horizontally. The best multi-column layouts use a majority of small columns with ratios like 1:1:4:2. Even 1:1:1:1 multi-column layouts can look very boring unless you are creative with them, but as with 3 column grids, you can play with overlapping and flow to create a design that is unique, useful, and fun.
